Histone deacetylase 8 regulates cortactin deacetylation and contraction in smooth muscle tissues.

چکیده
Histone deacetylases (HDACs) are a family of enzymes that mediate nucleosomal histone deacetylation and gene expression. Some members of the HDAC family have also been implicated in nonhistone protein deacetylation, which modulates cell-cycle control, differentiation, and cell migration. However, the role of HDACs in smooth muscle contraction is largely unknown. Here, HDAC8 was localized both in the cytoplasm and the nucleus of mouse and human smooth muscle cells. Knockdown of HDAC8 by lentivirus-encoding HDAC8 shRNA inhibited force development in response to acetylcholine. Treatment of smooth muscle tissues with HDAC8 inhibitor XXIV (OSU-HDAC-44) induced relaxation of precontracted smooth muscle tissues. In addition, cortactin is an actin-regulatory protein that undergoes deacetylation during migration of NIH 3T3 cells. In this study, acetylcholine stimulation induced cortactin deacetylation in mouse and human smooth muscle tissues, as evidenced by immunoblot analysis using antibody against acetylated lysine. Knockdown of HDAC8 by RNAi or treatment with the inhibitor attenuated cortactin deacetylation and actin polymerization without affecting myosin activation. Furthermore, expression of a charge-neutralizing cortactin mutant inhibited contraction and actin dynamics during contractile activation. These results suggest a novel mechanism for the regulation of smooth muscle contraction. In response to contractile stimulation, HDAC8 may mediate cortactin deacetylation, which subsequently promotes actin filament polymerization and smooth muscle contraction.

Reply to "Letter to the editor: 'KDAC and the regulation of nonnuclear smooth muscle protein acetylation'".
REPLY: We appreciate the interest expressed by Taggart and colleagues (9) in our article (6) for American Journal of Physiology-Cell Physiology. They have raised several issues regarding the paper. Below are detailed our responses. To assess the role of histone deacetylase 8 (HDAC8), we used lentivirus-mediated short hairpin RNA (shRNA) against HDAC8 as a major tool. Knockdown of HDAC8 attenuat...

TO THE EDITOR: We were intrigued to read the paper recently published by Li et al. (4) entitled “Histone deacetylase 8 regulates cortactin deacetylation and contraction in smooth muscle tissues.” The posttranslational modification of nonnuclear smooth muscle proteins by acetylation is emerging as an important regulatory mechanism, and our attention was drawn to several aspects of the article. F...
متن کاملClass IIb HDAC6 regulates endothelial cell migration and angiogenesis by deacetylation of cortactin.
Histone deacetylases (HDACs) deacetylate histones and non-histone proteins, thereby affecting protein activity and gene expression. The regulation and function of the cytoplasmic class IIb HDAC6 in endothelial cells (ECs) is largely unexplored. Here, we demonstrate that HDAC6 is upregulated by hypoxia and is essential for angiogenesis. Silencing of HDAC6 in ECs decreases sprouting and migration...
متن کاملDeacetylation of α-tubulin and cortactin is required for HDAC6 to trigger ciliary disassembly.
Cilia play important roles in sensing extracellular signals and directing fluid flow. Ciliary dysfunction is associated with a variety of diseases known as ciliopathies. Histone deacetylase 6 (HDAC6) has recently emerged as a major driver of ciliary disassembly, but little is known about the downstream players. Here we provide the first evidence that HDAC6-mediated deacetylation of α-tubulin an...
متن کاملHistone deacetylase 6 regulates cytokinesis and erythrocyte enucleation through deacetylation of formin protein mDia2
The formin protein mDia2 plays a critical role in a number of cellular processes through its ability to promote nucleation and elongation of actin filaments. In erythroblasts, this includes control of cytokinesis and enucleation by regulating contractile actin ring formation. Here we report a novel mechanism of how mDia2 is regulated: through acetylation and deacetylation at lysine 970 in the f...
